The county was established in 1823 and named after Revolutionary War hero Francis Marion. The county seat is Salem, and the largest city is Centralia. Marion County is located in the south-central part of Illinois and is bordered by Clinton County to the north, Jefferson County to the east, Washington County to the south, and Clay and Fayette Counties to the west. The county has a total area of 576 square miles, of which 574 square miles is land and 2 square miles (0.3%) is water. The terrain is mostly flat with some rolling hills. The Kaskaskia River flows through the county from north to south.
The county is divided into twelve townships: Centralia, East Salem, Farina, Iuka, Kinmundy, Meacham, Patoka, Raccoon, Romine, Sandoval, South Fork, and Walnut Hill.

As of the 2010 census, there were 39,437 people, 15,845 households, and 10,945 families residing in the county. The population density was 68.5 people per square mile. The racial makeup of the county was 97.2% White, 0.7% Black or African American, 0.3% Native American, 0.3% Asian, 0.2% from other races, and 1.3% from two or more races. 1.3% of the population were Hispanic or Latino of any race.
There were 15,845 households, out of which 30.2% had children under the age of 18 living with them, 53.2% were married couples living together, 10.7% had a female householder with no husband present, and 31.7% were non-families. 27.2% of all households were made up of individuals, and 12.7% had someone living alone who was 65 years of age or older. The average household size was 2.45 and the average family size was 2.97.
The median age in the county was 40.3 years. 24.2% of residents were under the age of 18; 8.2% were between the ages of 18 and 24; 23.3% were from 25 to 44; 27.2% were from 45 to 64; and 17.1% were 65 years of age or older. The gender makeup of the county was 48.7% male and 51.3% female.
